Introduction to penicillin
Since 1928, British bacteriologist Fleming first discovered the world’s first antibiotic – penicillin, has been a hundred years of history. Penicillin (Penicillin, or transliteration of penicillin), also known as penicillin G, peillin G, is a class of antibiotics containing penicillane, which can destroy the cell wall of bacteria and play a bactericidal role in the breeding period of bacterial cells. It is extracted from penicillium. The production process of penicillin G can be divided into two steps: strain fermentation and extraction and refining. ① Strain fermentation: the penicillium is inoculated on the solid medium to culture penicillium spores. Then, the seed culture medium was inoculated into the sterilized culture medium containing phenylacetic acid precursors in the fermenter, sterile air was injected, stirred, and culture fermented to obtain penicillin fermentation liquid. ② Extraction and refining: the penicillin fermentation liquid is cooled and filtered. In the filtrate extractor, butyl acetate was used for multistage countercurrent extraction to obtain butyl ester extract. The butyl ester extract was decolorized by activated carbon, then added to the salt agent and azeotropic distillation to obtain potassium penicillin G. Penicillin G sodium salt is prepared from penicillin G potassium salt by ion exchange resin (sodium type).

An important process in the production of penicillin is the extraction of penicillin from penicillin fermentation broth with organic solvents such as butyl acetate. Because the fermentation liquid contains complex substances such as protein, sugar and mycelium, the interface between the organic phase and the water phase is unclear during extraction, and it is an emulsifying zone of certain strength, which has a great influence on the yield of the finished product. To this end, demulsifier must be used to demulsify, eliminate emulsification phenomenon, and achieve rapid and effective two-phase separation. However, different demulsifiers have different demulsification ability on organic phase, and the performance of demulsifier directly affects the effect of two-phase separation
